I'm switching from a Windows desktop PC and Windows laptop to an iMac and a MacBook Pro. Like the Windows machines, both Macs will be connected via my home network with my Asus RT-AC68U router. I want to take either the Windows laptop hard drive or the Windows desktop hard drive and turn it into an NAS drive.

My RT-AC68U has a USB port, and it offers four USB applications: AiDisk, Media Server, Network Place (Samba) Share Service, and FTP Share Service. I'm guessing that I'd want to plug the NAS drive into the router's USB port and use AiDisk--is that right?

No, a NAS is a stand alone network device. Can be wired or wireless. What you mean is to use the USB port to connect an external drive enclosure. Then you can use your old hard drive to share files through the Asus router.
Oh, if you upgrade the router to the latest firmware you can format that USB drive with HFS to be more compatible with your shiny new Macs!
