I have a couple of clients that have their own local server. When a user is connected to the local network, I want to have the DNS point to a local IP address and not the WAN IP Address that external DNS does.

For example, DD-WRT has DNSMasq options.
address=/mail.mydomain.com/192.168.1.2
address=/mail.myotherdomain.com/192.168.1.2

Is this possible with AsusWRT-Merlin?

Excellent, thanks very much.

Now for a newbie question, can you point me in the direction of methods to upload files?

Enable SSH access in router's GUI.

Then either log into the router with an SSH client and create the files, or use WinSCP (hint: make sure you select the SCP protocol).

Can this be done using DNSFilter on the LAN page? With global mode set to router and with the exceptions listed at the bottom using up to 3 specified custom (local IP) DNS servers?

No. He doesn't want to change the DNS server, he want's to change the IP address returned for the given domains.
