I'm trying to connect an Asus RT-AC88U to a mobile hotspot for Internet access.

The hotspot supports USB 2.0 and 802.11ac up to 433 Mbps.

Which should be faster ?

USB connection from the router to the hotspot.

Connect the router to the hotspot through a wireless media bridge/extender via 5 GHz.​

The router''s USB can't be used for hotspot tethering.

Currently, the router running the current version of Asuswrt-Merlin is connected to the hotspot via USB and is working.​
 
What mobile hotspot are you using?

My apologies for my error.
Leave it alone. USB 3.0 will not limit your speed and will be more reliable than WiFi
 
I meant, what make/model of mobile hotspot. Might be nice to have as a backup solution for home.
 
Asus supports Android tethering, however your result might vary based on your actual device.

Wifi-based tethering (also known as "wireless client mode") is not supported.
