On the WAN settings page of AX88U pro you can set custom DNS with TLS. However there is no such option on the IPv6 tab.

Is this not supported?

I have a feature request out for it. I know I can use it as software in windows as well but would be neat to have it at router level.

Or can I add IPv6 in the IP fields of the WAN DoT?
 
You can add the IPv6 address on the WAN page. Here is what mine looks like (note: If you are running the current stock firmware for the AX88U Pro, the "Preset servers" box will not appear, but you can still manually specify both the IPv4 and IPv6 addresses, as well as the TLS Hostname):

After setting the IPv6 addresses in the DNS-over-TLS box on the WAN page, you will still need to specify the IPv6 DNS addresses on the IPv6 tab:

I feel it is a good idea to alternate the IPV4 and IPV6 DoT resolvers in the list.
